My First Successful Ligation

Every first-time is just as important, this applies to my first ligation, too! Click on the link to check what the hell does ligation mean. Now, after knowing what ligation is, you must be wondering what's the big deal about it - it's just putting the enzymes and different DNA fragment together in a tube. No BIG deal, you're right.

The catch is, the probably of successful ligation is soooooo darn low, so low that some people take THREE months (If your luck is so suay, it may be longer) to get it done. If you failed, it'll take you AT LEAST 3 days to PREPARE for the next experiment. It's very time consuming and tedious.

After numerous failures, you'll do all sorts of funny things like casting spells and drawing lucky charms to make sure your stuffs work.

And by the time you get it done properly, you need to transform the recombinant DNA into a new cell host. Easy as it sounds... The thing is, you wouldn't know whether your failure is caused by ligation or transformation untill you grow your newly transformed cells on agar plate - having colonies means there were ligations but it doesn't mean they are the correct ligation. This will take another day... (4 days till now)

If you're so lucky that you've colonies appear on your plate (assuming that your charms and spells work) then you'll extract the DNA out and test if they are the correct ones or not. This will take ANOTHER day... (total of 5 days) If you fail the test, you have to repeat the WHOLE THING AGAIN.

Woo hooo... Ligation is soooo FUN!

Hi folks! I'm Palastine, the retard standing on my right is Israel and I hate him.

Mom loves Israel more than me. She feeds him with sugar while stuffing me with thin air. We quarrel almost every single day for territory rights in our shelve but Mom always throw me into the back of Soy Sauce whenever Israel complaints to her.

But we learn how to live with it. We don't fight and we don't push each other down the shelve because we know we are part of the Kitchen. The kitchen needs us and we need the Kitchen.

Therefore, we learn to accommodate one another for the sake of the Kitchen.

Israeli Assault on Gaza

OK, Israeli assault on the Gaza is nothing new now. It has lasted for about 2 weeks since the first air strike. Now, let's see how U.S. reacts to the issue... Quoted from Channelnewsasia.

US President George W. Bush, in remarks to be broadcast on Saturday, urged all able parties to press Hamas to stop firing rockets at Israel and secure a lasting ceasefire, after a week of heavy Israeli air strikes on the Gaza Strip.

His administration meanwhile gave Israel free rein over whether to send ground troops into Gaza, despite growing criticism over its handling of a conflict that has killed at least 436 Palestinians and left 2,290 others wounded...

...Bush said the Israeli strikes were in self-defense after Hamas let a six-month ceasefire lapse on December 19 and fired rockets at Israel...

Zbigniew Brzezinski, who was national security adviser for US president Jimmy Carter, told CNN television that Rice's remarks "clearly show that the US policy right now is completely bankrupt" and the Israeli offensive "will further radicalise the Palestinians"...

...But the US president-elect has kept silent on the latest phase of the Israeli-Palestinian conflict, with his aides saying their boss was "monitoring" the situation.

So, it's like a child's game la. I slap your face, then you slap me back. I wanna have a greater revenge, I ask my big brother to slap you again. The Little Red Dot

If you wonder why people call Singapore as the Little Red Dot and how it came about... Read on, if you are interested.

When Bacharuddin Jusuf Habibie was the president of Indonesia 10 years ago (1998-1999), he named Singapore as the Little Red Dot on map in the Asian Wall Street Journal dated 4th August 1998. This is what he said:

It was reported that Habibie had remarked that he did not have the feeling that Singapore was a friend and had pointed to a map, saying: "It's OK with me, but there are 211 million people (in Indonesia). All the green (area) is Indonesia and that red dot is Singapore."

What's more interesting is how the then Singapore Prime Minister, Goh Chok Tong replied.19 days after Habibie remarked in Asian Wall Street Journal on the National Day Rally Speech. Back then, many countries in this region was striked by the Asian financial crisis and Indonesia was particularly hit hard. Goh said:

"Singapore will help Indonesia within the limits of our ability. We are a small economy. ... After all we are only three million people. Just a little red dot on the map. Where is the capacity to help 211 million people?"

War of words between leaders of nations. We have tonnes of those in the Bolehland, too. Dr. M is one of them. =) Cheers.
