My First Successful Ligation

Every first-time is just as important, this applies to my first ligation, too! Click on the link to check what the hell does ligation mean. Now, after knowing what ligation is, you must be wondering what's the big deal about it - it's just putting the enzymes and different DNA fragment together in a tube. No BIG deal, you're right.

The catch is, the probably of successful ligation is soooooo darn low, so low that some people take THREE months (If your luck is so suay, it may be longer) to get it done. If you failed, it'll take you AT LEAST 3 days to PREPARE for the next experiment. It's very time consuming and tedious.

After numerous failures, you'll do all sorts of funny things like casting spells and drawing lucky charms to make sure your stuffs work.

And by the time you get it done properly, you need to transform the recombinant DNA into a new cell host. Easy as it sounds... The thing is, you wouldn't know whether your failure is caused by ligation or transformation untill you grow your newly transformed cells on agar plate - having colonies means there were ligations but it doesn't mean they are the correct ligation. This will take another day... (4 days till now)

If you're so lucky that you've colonies appear on your plate (assuming that your charms and spells work) then you'll extract the DNA out and test if they are the correct ones or not. This will take ANOTHER day... (total of 5 days) If you fail the test, you have to repeat the WHOLE THING AGAIN.

Woo hooo... Ligation is soooo FUN!

The Minus -Eighty-Degree-Celsius Freezer

There is a minus-eighty-degree-Celsius freezer hiding in a corner of NTU. (OK, its at NTU-SCBE-CBE-B3-30. Come and steal it.)

Mind you, if you are thinking of stealing this baby, remember to wear a piece of high-quality hand glove. Moboto.blogspot.com shall not be responsible if your hand got deep froze and detaches itself from your arm.

Behold, it's the all mighty minus-eighty-degree-Celsius freezer!

I know this is a darn outdated entry... but who cares~

We can't afford to buy a real pear tree, neither an ordinary fake pear tree.
But it's okay, we have a micro version.

So-long-ago Farewell Party

Yeap yeap, it's a long-ago-picture from Aug 22, 2008, but come on folks! I am a freaking busy man, you know! It was Rachael's farewell party after working in Singapore for two months - time really flies like flies huh?

The Longbar is the birth place of Singapore Sling and first served in 1915. What is this Singapore-sling-around thing? The ingredients are gin, Cointreau, cherry brandy, Dom Benedictine, pineapple juice, Grenadine, Angoustura bitters and limes. However, I suggest having some beer before drinking the sling, or rather, try to mix a little of soda into the drink. It'll definitely taste better that way.
Yeap, we chilled out at the Long Bar, tried out Singapore Sling (the Long Bar invented it), eating peanuts, throwing peanuts shells anywhere you like and I love it :) What to do? After all, I am a Malaysian who like to throw rubbish everywhere and anywhere.

And we went to Zouk out after that...
Yeah yeah, I know I am very zombie that night...
